Abstract


Abstract: This study aims to determine the right buffer for use in DNA genomic isolation of the Sphagnum sp. and Stenochlaena palustris ferns and produce handout for class XII Senior high school. This research was conducted at the Genetic Laboratory of the Department of Biology, Faculty of Mathematics and Natural Sciences, University of Riau in March-June 2019. The parameters of this study were the presence of DNA in the supernatant, DNA band thickness and DNA concentration based on electrophoresis results. Handout design is carried out using 2 stages, analysis and design. The results showed that the Sphagnum sp. and Stenochlaena palustris ferns successfully isolated using detergent buffer and CTAB both using liquid nitrogen and without liquid nitrogen, which is marked by the appearance of lumps or white fibers in the supernatant layer. While samples using liquid soap were not successful. Therefore, detergent is more appropriate to be used for simple DNA genomic isolation experiments in senior high school. Genomic DNA based on electrophoresis results was obtained in 7 samples from a total of 12 samples, which are indicated by the presence of DNA bands. The highest DNA concentration was found in Stenochlaena palustris fern sample with detergent buffer and CTAB and Sphagnum sp. sample with CTAB buffer that was 50 ng/μl, while the lowest DNA concentration was found in Stenochlaena palustris sample with liquid soap buffer which was 15 ng/μl. The results of the research obtained will be used as a development of learning genetic material for Class XII senior High School in KD 3.3 and 4.3. The results of the design of learning tools in the form of syllabus, lesson plans and handout.
